牛耳枫抗胃溃疡和抗炎活性部位比较研究
Comparison on the Daphniphyllum calycinum Benth.Extracts with Anti-Gastric Ulcer and Anti-Inflammatory Activities

Abstract
Objective:To compare the Daphniphyllum calycinum Benth.ethyl acetate extract(DC-EA)and dichloromethane extract(DC-DCM)with anti-gastric ulcer and anti-inflammatory activities,and explore their gastroprotective mechanisms.Methods:HPLC was employed to ana-lyze the chemical compounds in DC-EA and DC-DCM.The ethanol-induced gastric ulcer model,the lipopolysaccharide(LPS)-induced in-flammation model of RAW264.7 macrophages,and the mouse model of endotoxemia were established.After treatment with DC-EA and DC-DCM,the gastric ulcer area was measured by Image J,and the inhibition rate on gastric ulcer was calculated.The histological changes and mucoprotein distribution were observed by hematoxylin-eosin staining and periodic acid-Schiff(PAS)staining.The levels of tumor necrosis factor(TNF)-α and interleukin(IL)-1β in the serum and cell culture supernatant as well as malondialdehyde(MDA)and glutathione(GSH)in the gastric tissue were measured by ELISA.The levels of nitric oxide(NO)in the serum and cell culture supernatant were deter-mined by Griess.The analysis of variance and the technique for order preference by similarity to an ideal solution(TOPSIS)were employed to screen the extract with anti-inflammatory activity.Results:The content of six compounds in DC-EA and only 1 compound in DC-DCM was determined.Compared with the normal control group,the ethanol-induced gastric ulcer model showed obvious damage in the gastric tissue,in-creased ulcer area(P<0.01),elevated levels of TNF-α and IL-1β in the serum and MDA in the gastric tissue(P<0.01),decreased PAS-positive area,and lowered GSH level in the gastric tissue(P<0.01).Compared with the model group,DC-EA(2 g/kg and 4 g/kg)allevia-ted the damage of the gastric tissue,reduced the ulcer area(P<0.01),lowered the levels of TNF-α and IL-1β in the serum and the MDA lev-el in the gastric tissue(P<0.01,P<0.05),increased the PAS-positive area,and elevated the GSH level in the gastric tissue(P<0.01).Compared with the normal control group,the inflammation models in vitro and in vivo showed elevated levels of NO,TNF-α,and IL-1β in the cell culture supernatant and serum(P<0.01).Compared with the model control group,DC-EA at 25,50,and 100 mg/mL and 2 g/kg and 4 g/kg inhibited the release of NO,TNF-α,and IL-1β(P<0.01).Moreover,DC-EA(2 g/kg and 4 g/kg)showed stronger anti-inflammatory effect than DC-DCM.Conclusion:DC-EA has gastroprotective and anti-inflammatory activities,and it may exert the gastroprotective effect by lowering the levels of inflammatory cytokines and oxidative stress.关键词
