代代花黄酮类成分指纹图谱研究及8个成分含量测定
Study on Fingerprint of Flavonoids in the Flower of Citrus aurantium var.amara and Determination of the Contents of 8 Components
摘要
Abstract
Objective:To establish the HPLC fingerprint of the flower of Citrus aurantium var.amara and to determine the contents of 8 flavonoids narirutin,naringin,hesperidin,neohesperidin,naringenin,hesperidin,nobiletin and tangeretin.Methods:Using Diamonsil(2)C18(250 mm×4.6 mm,5 μm)chromatographic column,the mobile phase was acetonitrile-0.1%phosphoric acid solution with gra-dient elution.The flow rate was 1.0 mL/min,detection wavelength was 320 nm,column temperature was 30℃,sample size was 10 μL.The similarity evaluation,cluster analysis(CA),principal component analysis(PCA)and orthogonal partial least square discriminant a-nalysis(OPLS-DA)of 21 batches of samples from different origin were carried out to select the quality difference components.Results:The similarity of fingerprints of 21 batches of flower of Citrus aurantium var.amara was 0.983~0.999,19 common peaks were identi-fied,8 components were identified by reference products,21 batches of flower of Citrus aurantium var.amara were divided into 2 catego-ries by CA and PCA,OPLS-DA screened out 7 components that caused the difference in the classification of Citrus aurantium var.amara flower from different places.The contents of above eight components were 2.30~10.00 mg/g,27.25~60.87 mg/g,5.61~16.31 mg/g,44.07~84.17 mg/g,0.03~0.78 mg/g,0.02~0.88 mg/g,0.05~0.14 mg/g,0.11~0.24 mg/g,respectively.Conclu-sion:The fingerprint and multi-index content determination method established in this study is accurate and reliable,and can provide a basis for the quality evaluation of Citrus aurantium var.amara flower.关键词
