川芎嗪调节Wnt/β-catenin信号通路对早发性卵巢功能不全大鼠卵巢颗粒细胞凋亡的影响
Effect of Tetramethyipyrazine on Apoptosis of Ovarian Granulosa Cells in Rats with Premature Ovarian Insufficiency by Regulating Wnt/β-catenin Signaling Pathway

Abstract
Objective:To investigate the effect of tetramethyipyrazine on apoptosis of ovarian granulosa cells in rats with premature ovarian insufficiency(POI)by regulating Wnt/β-catenin signaling pathway.Methods:SD rats were randomly divided into blank control group,model group,tetramethyipyrazine low,medium and high-dose groups,high dose tetramethyipyrazine with XAV-939(Wnt/β-cate-nin signaling pathway inhibitors)group.Except for the blank control group,the rats in other groups were intraperitoneally injected with cyclophosphamide to construct POI model.The estrus cycle of rats in each group was observed.Body mass and ovarian index were meas-ured.ELISA was used to detect serum FSH,E2,LH and AMH levels.HE staining was applied to observe pathological changes in ovarian tissue and the follicles at all levels were counted.TUNEL staining was applied to observe apoptosis of ovarian granulosa cells.Immuno-histochemistry was applied to observe the expressions of Bcl-2 and β-catenin.Western Blot was applied to detect the expressions of Bcl-2,Bax,β-catenin and Cyclin D1 proteins in ovarian tissue.Results:Compared with blank control group,the estrous cycle of rats in model group was disturbed,ovarian atrophy,body mass,ovarian index,E2,AMH,normal follicle number,Bcl-2,β-catenin,Cyclin D1 expres-sions were significantly decreased,FSH,LH,atretic follicle number,granulosa cell apoptosis rate and Bax protein expression were signif-icantly increased(P<0.05).Compared with model group,body mass,ovarian index,E2,AMH,normal follicle number and protein ex-pressions of Bcl-2,β-catenin and Cyclin D1 in rats were significantly increased in each tetramethyipyrazine group,while the rate of es-trous cycle disturbance,FSH,LH,atretic follicle number,granulosa cell apoptosis rate and Bax protein expression were significantly de-creased(P<0.05).XAV-939 could reverse the effect of tetramethyipyrazine(P<0.05).Conclusion:Tetramethyipyrazine can improve o-varian function and reduce ovarian granulosa cell apoptosis in POI rats,the mechanism may be related to activation of Wnt/β-catenin signaling pathway.关键词
