摘要

Abstract

[Objective]To explore the genetic diversity and genetic relationship of 15 Carum carvi germplasms in Qinghai region,and to pro-vide theoretical support for the artificial cultivation of C.carvi varieties.[Method]DNA from 149 fresh leaves of C.carvi was extracted by modi-fied CTAB method,and then 10 high-efficiency ISSR primers were used for molecular labeling.[Result]A total of 8 412 polymorphic bands were obtained,the total polymorphism ratio was 90.21%,the average Nei's gene diversity index(H′)was 0.50,the average Shannon's infor-mation index(I)was 0.75,and the genetic similarity coefficient among C.carvi germplasm was 0.84-0.97.The genetic diversity index(Ht),intra-population genetic diversity index(Hs),gene differentiation coefficient(Gst)and gene flow(Nm)were 0.23,0.17,0.26 and 1.42,re-spectively.The 15 germplasms were divided into three categories.The 149 C.carvi individuals were divided into 6 categories.[Conclusion]The germplasm of wild C.carvi in Qinghai area has rich genetic diversity,and there is extensive gene exchange between germplasms,and this gene exchange shows a certain gradual diffusion law in the same direction in the region,and the rich genetic diversity of wild C.carvi in Qinghai area comes from the differences between individuals.
