灰葡萄孢致病蛋白BcPDR1的功能区域鉴定
Identification of the functional region of the pathogenic protein BcPDR1 in Botrytis cinerea

Abstract
Botrytis cinerea is a typical necrotrophic plant pathogenic fungus that can damage thousands of plant species and cause gray mold.Previous studies conducted in our laboratory identified a novel pathogenic gene,BcPDR1(Pathogenicity Deficient Related 1),which positively regulates the growth,development,and pathogenicity of B.cinerea.However,bioinformatics analyses did not reveal any homologous genes or conserved domains associated with BcPDR1.To investigate the mechanism of this gene,we analyzed the important functional regions of BcPDR1 in this study.The BcPDR1 protein was divided into three regions:BcPDR1-1(1~80 amino acids),BcPDR1-2(81~121 amino acids),and BcPDR1-3(122~199 amino acids).Using the BcPDR1 gene knockout mutant ΔBcpdr1,we constructed three reverse mutants through a gene complementation recovery technique.We then analyzed the phenotypes and pathogenicity of these three regional reversion mutants.The results showed that the colony morphology,mycelial morphology,growth rate,and infection structures of the three regional reversion mutants were largely consistent with those of the ΔBcpdr1 mutant.Notably,significant disease spots were observed on tomato fruits and tobacco leaves,with the lesion produced by the reversion mutant BcPDR1-3 being the most pronounced.This finding suggested that BcPDR1-3(122~199 amino acids)was the crucial functional region of the BcPDR1 protein.These results provide a solid foundation for elucidating the molecular mechanisms by which BcPDR1 regulates the growth and development of pathogens,as well as for the development of targeted drugs for the prevention and treatment of gray mold.关键词
