Function of transcription factor VdbZIP2 in Verticillium dahliae,pathogen of verticillium wilt of smoke tree

Abstract
To deeply investigate the pathogenic mechanism of Verticillium dahliae,the causal agent of the devastating forest disease verticillium wilt of smoke tree,VdbZIP2,a bZIP transcription factor gene was deleted by PEG-mediated protoplast genetic transformation method.The biological traits of the mutants were analyzed,and the pathogenicity was determined.The results showed that knockout of VdbZIP2 seriously affected melanin synthesis,microsclerotia formation,cell wall stress sensitivity,and available nitrogen source utilization in V.dahliae.However,the mutants were fully pathogenic,indicating VdbZIP2 is not a virulence-essential gene.In summary,we conclude that the transcription factor VdbZIP2 of V.dahliae is involved in microsclerotia formation,adversity stress and nitrogen source utilization,which provides important clues for the subsequent in-depth analysis of the molecular regulatory network of VdbZIP2.关键词
