益气开秘方介导VIP/cAMP/PKA/AQP信号通路治疗衰老模型大鼠慢传输便秘的作用机制
Exploring the mechanism of Yiqi Kaimi Formula in treating slow transit constipation in aging model rats via the VIP/cAMP/PKA/AQP signaling pathway

Abstract
Objective To explore the mechanism by which Yiqi Kaimi Formula(YQKMF)treats slow transit constipation(STC)in ag-ing model rats with both qi and yin deficiency syndrome.Methods An aging STC rat model was established.After the experiment,fecal water content and intestinal ink progradation rate were measured;colon tissue pathology was examined;serum levels of vasoactive intesti-nal peptide(VIP),superoxide dismutase(SOD),and catalase(CAT)were detected.The mRNA and protein expression levels of pro-tein kinase A(PKA),aquaporin-1(AQP1),and aquaporin-9(AQP9)in the colon tissues were also assessed.Results Compared with the blank group,the model group showed reduced fecal water content,decreased intestinal ink progradation rate,a reduction in gob-let cells,and increased inflammatory cells in the colon tissues.The expression of phosphorylated PKA(p-PKA),as well as PKA and AQP9 mRNA and protein were decreased,while AQP1 mRNA and protein expression were increased.Serum levels of VIP,SOD,and CAT were also reduced(P<0.05).Compared with the model group,the Mosapride and YQKMF groups showed increased fecal water content and enhanced intestinal ink progradation rate.The number of goblet cells were increased,and inflammatory factors decreased.Additionally,p-PKA expression was elevated,PKA and AQP9 mRNA and protein expression increased,while AQP1 mRNA and pro-tein expression decreased.Serum VIP,SOD,and CAT levels were also elevated(P<0.05).Conclusion YQKMF can alleviate consti-pation symptoms in aging model rats with STC by modulating the VIP/cAMP/PKA/AQP signaling pathway.关键词
