摘要

Abstract

Objective To investigate the effects of Qianggu Yin(QGY),/Bone-strengthening Decoction a traditional Chinese medicine(TCM)compound formula,on bone microstructure in osteoporosis(OP)and to explore its mechanism of regulating OPG/RANKL/RANK by intervening complements of C3 and SIRT5.Methods A postmenopausal OP mouse model was established via bilateral ovariec-tomy.Mice were randomly assigned to the sham group,the model group,or the QGY group.The QGY group received QGY via gavage,while the sham and model groups received an equal volume of normal saline.After eight weeks of treatment,the mice were sacrificed,and femoral tissues were collected for TRAP staining to assess osteoclast differentiation.Micro-CT and RT-qPCR were used to evaluate trabecular number,trabecular bone volume fraction,trabecular thickness,and trabecular spacing.Additionally,QGY-containing serum and control serum were prepared.The expression levels of C3,SIRT5,osteoprotegerin(OPG),and receptor activator of nuclear factor-κB ligand(RANKL)during osteoblast and osteoclast differentiation were assessed using qRT-PCR and Western blot(WB).Re-sults Compared with the sham group and model group,bone microstructure of mice in the QGY group was significantly improved.Addi-tionally,QGY inhibited complement C3,affected the succinylation key protein SIRT 5,and alleviated OP.In cell experiments,inhibited C3 gene expression was found to up-regulate the OPG expression,promote the expression of SIRT 5,a key protein for succinylation,and reduce the expression of RANKL.Conclusion QGY may exert its therapeutic effect on OP by modulating C3 levels and the succi-nylation-related protein SIRT5,thereby regulating OPG and RANKL expression.
