黄芪赤风汤对脑缺血/再灌注损伤的神经保护作用及对小鼠血清中CRP、TNF-α、IL-18、IL-1β表达的影响
Neuroprotective effects of Huangqi Chifeng Decoction on cerebral ischemia/reperfusion injury and its impact on the expressions of CRP,TNF-α,IL-18 and IL-1β in mouse serum

Abstract
Objective To investigate the neuroprotective effects of Huangqi Chifeng Decoction(HQCFD)on cerebral ischemia/reperfu-sion(I/R)injury and its influence on the expression of inflammatory markers CRP,TNF-α,IL-18,and IL-1β in mouse serum.Methods In vitro,BV2 cells were divided into five groups:the control group,model group,and three HQCFD-treated groups at low,medium,and high doses.An oxygen-glucose deprivation/reoxygenation(OGD/R)model was established,and cell viability was as-sessed using the CCK-8 assay.In vivo,twenty-five C57BL/6 mice were randomly assigned to five groups:the sham group,model group,and HQCFD-treated groups at low,medium,and high doses.A middle cerebral artery occlusion/reperfusion(MCAO/R)model was established.Neurological function was evaluated using the modified neurological severity score(mNSS).TTC staining was employed to assess brain infarct volume,while HE and Nissl staining were used to observe neuronal changes in the brain tissues.Serum levels of CRP,TNF-α,IL-18,and IL-1β were measured by ELISA.Results In vitro,the survival rate of BV2 cells in the model group was significantly lower than that in the control group(P=0.0004<0.001).BV2 cell viability were improved in all HQCFD-treated groups,with the high-dose group showing the most prominent effect;however,the difference was not statistically significant(P=0.3459>0.05).In vivo,the mNSS scores in the model group were significantly higher than those in the sham group(P<0.0001),accompanied by a marked increase in brain infarct volume((P=0.0002<0.001).Neuronal degeneration,necrosis,vacuo-lation,nuclear condensation,deep staining,cytoplasmic Nissl body reduction,and pale staining were observed in the infarct area.Ser-um levels of CRP,TNF-α,IL-18,and IL-1β were significantly elevated(P<0.0001).Compared to the model group,mNSS scores were decreased in all HQCFD-treated groups,with the high-dose group showing the most pronounced reduction(P=0.0002<0.001).Brain infarct volume was also decreased,especially in the high-dose group(P=0.0144<0.05).Neuronal morphologi-cal conditions in the infarct area were improved,and serum levels of CRP,TNF-α,IL-18,and IL-1β decreased,with the greatest reduction observed in the high-dose group((P=0.0001<0.001;(P=0.0011<0.01;(P<0.0001;(P=0.0003<0.001).Conclusion HQCFD exerts neuroprotective effects against cerebral I/R injury,which may be closely associated with the modu-lation of neuroinflammation mediated by inflammatory markers such as CRP,IL-18,and IL-1β.关键词
