基于NF-κB/COX-2通路探讨祛瘀温经汤治疗寒凝血瘀型原发性痛经的作用机制
Investigating the mechanism of Quyu Wenjing Decoction in alleviating primary dysmenorrhea with cold coagulation and blood stasis via the NF-κB/COX-2 pathway

Abstract
Objective To investigate the therapeutic effects and underlying mechanism of Quyu Wenjing Decoction(QYWJD)on prima-ry dysmenorrhea(PD)with cold coagulation and blood stasis in rats based on the nuclear factor-kappa B(NF-κB)/cyclooxygenase-2(COX-2)signaling pathway.Methods Sixty female Sprague-Dawley(SD)rats were randomly assigned to the following groups:the blank group,model group,the QYWJD low-,medium-,and high-dose groups,and the Shaofu Zhuyu Granule(SFZYG)group.After the drug intervention,the writhing response and its latency were observed.Hemorheological parameters were measured,and hema-toxylin-eosin(HE)staining was performed to observe uterine tissue pathology.Cytokine levels were determined using enzyme-linked immunosorbent assay(ELISA),while NF-κB and COX-2 protein expression levels were assessed by Western blot(WB).The mRNA expression levels of NF-κB and COX-2 were detected via real-time PCR.Results Compared with the blank group,rats in the model group exhibited significant writhing responses,increased infiltration of inflammatory cells in uterine tissue,elevated levels of IL-6,TNF-α,and PGF2α,reduced PGE2 levels,and upregulated NF-κB and COX-2 protein and mRNA expression(P<0.05).Compared with the model group,the QYWJD-treated groups demonstrated prolonged latency of writhing response,fewer writhing episodes,re-duced inflammatory cell infiltration in the uterine tissues,decreased IL-6,TNF-α,and PGF2α levels,increased PGE2 levels,and reduced NF-κB and COX-2 and mRNA expression(P<0.05).Conclusion QYWJD may alleviate PD with cold coagulation and blood stasis by modulating the NF-κB/COX-2 pathway,inhibiting the expression of inflammation-related proteins,and repairing the uterine tissue damage.关键词
