复方甲亢片对GD模型大鼠甲状腺滤泡上皮细胞TSH受体完整性的保护作用
Protective effects of Fufang Jiakang Tablet on TSH receptor integrity in thyroid follicular epi-thelial cells of model rats with Graves'disease

Abstract
Objective To investigate the protective effects of Fufang Jiakang Tablet(FJT)on the integrity of thyroid-stimulating hor-mone receptor(TSHR)in thyroid follicular epithelial cells of Graves'disease(GD)model rats and to explore its potential mechanism in reducing TSHreceptorantibody(TRAb)levels.Methods Fortyfemale Sprague-Dawley(SD)rats(8 weeks old)were randomly as-signed to four groups:the normal control group,model group,FJT group,and methimazole group,with 10 rats in each group.Experi-mental GD models were established via intravenous immunization with Yersinia enterocolitica.The FJT group received 0.47 g/kg of FJT suspension via gavage,while the methimazole group received 1.58 mg/kg of methimazole suspension.The normal control and model groups were administered an equal volume of saline.All interventions lasted for 30 days.Serum levels of free triiodothyronine(FT3),free thyroxine(FT4),thyroid-stimulating hormone(TSH),and TRAb were measured using ELISA.The relative expression of the TSHR A subunit in thyroid follicular epithelial cells was assessed by immunofluorescence assay.Results Compared with the normal con-trol group,the model group exhibited significantly increased serum FT3 and FT4 levels,decreased TSH levels,elevated TRAb titers,and reduced relative expression of the TSHR A subunit in thyroid follicular epithelial cells(all P<0.01).Compared with the model group,both the FJT and methimazole groups showed reduced FT3 and FT4 levels,increased TSH levels,decreased TRAb titers,and in-creased TSHR A subunit expression(all P<0.01).Compared with the methimazole group,the FJT group demonstrated greater reduc-tions in FT3 and FT4 levels,a more significant increase in TSH levels,a greater decrease in TRAb titers,and a more pronounced in-crease in TSHR A subunit expression(P<0.05).Conclusion FJT may protect the integrity of TSHR in GD model rats by reducing the shedding of TSHR A subunits,thereby lowering the TRAb levels and contributing to better GD management.关键词
