Investigating the mechanism of ferroptosis induced by bacillus coagulans MZY531 via the Wnt/β-catenin signaling pathway in human hepatocellular carcinoma cells

Abstract
Objective To investigate the mechanism by which Bacillus coagulans MZY531 induces ferroptosis in human hepatocellular carcinoma(HCC)cells through the Wnt/β-catenin signaling pathway.Methods The effect of B.coagulans MZY531 on the prolifera-tion of HepG2 and SK-Hep-1 HCC cells was assessed using the CCK-8 assay.The levels of malondialdehyde(MDA)and glutathi-one(GSH)in HCC cells were measured by the micro-colorimetric method.Intracellular Fe2+and lipid peroxide levels were detected u-sing a fluorescent probe assay.Western blot(WB)was performed to assess the expression of key ferroptosis-related proteins and Wnt/β-catenin pathway-associated proteins.Ferroptosis inhibitor Ferrostatin-1 and pathway inhibitor IWR-1 were used to further explore the intervention of the Wnt/β-catenin pathway in B.coagulans MZY531-induced ferroptosis.Results MZY531 inhibited the prolifera-tion of HCC cells,significantly increased intracellular Fe2+and lipid peroxide levels,elevated MDA expression,suppressed GSH expres-sion,and downregulated the expression of key ferroptosis-related proteins and Wnt/β-catenin pathway-associated proteins.Following the co-treatment with MZY531 and Ferrostatin-1,ferroptosis was markedly induced in HCC cells.Compared with the IWR-1 group,the MZY531+IWR-1 group exhibited further suppression of Wnt/β-catenin pathway-related proteins β-catenin,Cyclin D1,and c-Myc,along with downregulation of the ferroptosis-associated proteins GPX4 and SLC7A11.Conclusion B.coagulans MZY531 may induce ferroptosis in HCC cells by regulating the Wnt/β-catenin signaling pathway.关键词
