全真一气汤调控PIK3R3、PLA2G10、TGFBR1、BIRC5的表达水平抑制COPD向肺癌发展的机制研究

Abstract
Objective To observe the effects of Quanzhen Yiqi Decoction(全真一气汤,QZYQD)on the expression levels of PIK3R3,PLA2G10,TGFBR1,and BIRC5 in the lung tissues of rats with chronic obstructive pulmonary disease(COPD)and explore its impact on the incidence of lung cancer.Methods Eighteen Sprague-Dawley(SD)rats were randomly divided into the Chinese herbal medicine(CHM)group and model group,with 9 rats in each group.A COPD rat model was established by sumdging combined with lipopolysac-charide(LPS).The model group was gavaged with daily intragastric administration of 0.9%normal saline(NS),while the CHM group was gavaged with daily intragastric administration of 40%concentrated QZYQD.PCR was used to detect mRNA expression levels of PIK3R3,PLA2G10,TGFBR1,and BIRC5,and Western blot(WB)was performed to measure protein levels in the lung tissues of rats.Results The mRNA expression levels of PIK3R3,PLA2G10,and BIRC5 in CHM group were lower than those in the model group,with expression abundances of 0.159-fold,0.138-fold,and 0.647-fold of the model group,respectively.The mRNA expression level of TGFBR1 in CHM group was higher,at 1.867-fold of the model group.Protein expressions of PIK3R3,PLA2G10 and BIRC5 were down-regulated in CHM group compared to the model group,while TGFBR1 protein expression was up-regulated.Conclusion-QZYQD may inhibit the onset of lung cancer in COPD by modulating the expressions of PIK3R3,PLA2G10,BIRC5 and TGFBR1.关键词
