百杏润肺化浊解毒汤对哮喘模型小鼠气道炎症的影响
Exploring the effect of Baixing Runfei Huazhuo Jiedu Decoction(百杏润肺化浊解毒汤)on airway inflammation in asthmatic model rats

Abstract
Objective To investigate the effect of Baixing Runfei Huazhuo Jiedu Decoction(百杏润肺化浊解毒汤,BRHJD)on the airway inflammation and related serum cytokines in ovalbumin(OVA)-induced asthmatic model mice.Methods Sixty BALB/c mice were randomly divided into six groups:the normal group,model group,montelukast sodium group,and BRHJD low/medium/high-dose groups,with 10 mice in each group.Except for the normal group replaced with 0.9%sodium chloride solution,all other groups were sensitized with OVA and stimulated with nasal drops to induce the establishment of an asthma model.All BRHJD groups were adminis-tered via gavage at doses of 12.95,25.90,and51.80 g·kg-1·d-1,respectively;the montelukast sodium group received 1.3 mg·kg-1·d-1 by gavage;while the normal and model groups were given 0.2 mL of 0.9%sodium chloride solution by gavage.All treat-ments were administered once daily for 7 consecutive days before stimulation by dripping nose.The pathological changes in airway inflam-mation in the lung tissues of mice were observed by Hematoxylin-eosin(HE)staining.The airway goblet cell hyperplasia and mucus secretion in mice were observed by periodic acid-Schiff(PAS)staining.Content of serum interleukin(IL)-6,IL-13 and tumor nec-rosis factor(TNF)-α were detected by enzyme-linked immunosorbent assay(ELISA).Results Compared with the normal group,the model group showed significant airway inflammation,goblet cell metaplasia,mucus hypersecretion,and increased contents of serum IL-6,IL-13,and TNF-α(P<0.01).Compared with the model group,all BRHJD dose groups and the montelukast sodium group ex-hibited reduced airway inflammation,mucus secretion,and goblet cell metaplasia in varied degrees.IL-6,IL-13,and TNF-α con-tents were decreased in the BRHJD high-dose and montelukast sodium groups(P<0.05 or P<0.01).The BRHJD low-dose group showed reduced contents of serum IL-6 and IL-13(P<0.05 or P<0.01),while the medium-dose group showed reduced contents of serum IL-13(P<0.01).Among them,the BRHJD medium-dose group exhibited higher serum IL-6 contents compared to the montelukast sodium and BRHJD high-dose groups(P<0.05).Conclusion BRHJD may alleviate the asthmatic symptoms by reducing the expression levels of cytokines to further reduce cytokines-mediated airway inflammation and mucus hypersecretion.关键词
