Abstract

Porcine pseudorabies is a highly contagious disease of pigs caused by the pseudorabies virus(PRV).It has the potential to cause death in young pigs and abortion in pregnant sows,resulting in significant economic losses for the pig farming industry.In addition to the typical clinical symptoms,PRV can also cause immunosuppression in pig populations,leading to secondary infections,increased treatment costs,and a reduction in feed intake and feed conversion efficiency.Furthermore,there is an increase in feed wastage and a delay in marketing.In order to reduce the economic losses incurred by the pig farming industry in Xinjiang,this study conducted research on the detection of pseudorabies virus,thereby providing the basic data required for the control of pseudorabies.Tissues from the brains,lungs,and lymph nodes of deceased lactating and weaned piglets were obtained from Xinjiang a certain pig farm and subjected to total DNA extraction.The gE gene of pseudorabies virus was identified using primers based on the national standard GB/T 18641-2018 Diagnostic method for pseudorabies,and the amplified product was 368 bp in size.The cloned and identified product was sent for sequencing,and the sequence analysis demonstrated that the PRV-gE gene sequence exhibited an identity of 97.7%-97.9%with foreign PRV-gE gene sequences and 99.4%-99.9%with domestic PRV-gE gene sequences.This indicated that the PRV-gE gene sequence of the pseudorabies virus strain was a Chinese prevalent strain.The findings of this study provide a reference point for the eradication of pseudorabies and the selection of an appropriate vaccine.
