Abstract

In this experiment,tissue culture was conducted on the leaves of Begonia maculata Raddi,and the effect of different disinfection time on the sterilization effect of the explants was studied.Effects of different concentrations of hormones on adventitious bud induction in the first generation,adventitious bud proliferation in subsequent generations and adventitious root induction in Begonia maculata were experimented.The effects of different substrate ratio on transplanting in order to establish the system of rapid propagation technique of Begonia maculata Raddi in tissue culture.The results showed that the survival rate of leaves was 100%after disinfection with 75%ethanol for 30 s and 2%sodium hypochlorite solution for 5 min.MS+1.0 mg/L 6-BA+0.6 mg/L IBA was suitable for adventitious bud induction,and the induction rate was 100%.MS+0.1 mg/L 6-BA+1.0 mg/L IBA was suitable for adventitious bud proliferation and propagation,and the proliferation coefficient was 1.40.Then the 1/2MS+0.2 mg/L NAA was suitable for adventitious root growth,and 1/2MS+0.2 mg/L IBA was suitable for adventitious root elongation,both rooting rates were 100%.The survival rate of the substrate suitable for acclimation and transplanting was 100%on the conditions of turf∶vermiculite=1∶1.
