Abstract

Objective:To investigate the therapeutic effects and mechanisms of electroacupuncture on a rat model of chronic obstructive pulmonary disease(COPD).Methods:Forty Sprague-Dawley(SD)rats were randomly divided into four groups:control,model,medication,and electroacupuncture,with 10 rats in each group.The model group was in-duced with COPD by repeated intratracheal instillation of lipopolysaccharide(LPS),while the control group received saline instead.The control group received no treatment,the medication group was treated with dexamethasone(5 mg/kg),and the electroacupuncture group received treatment at the bilateral BL13(Feishu)and ST36(Zusanli)acupoints with an electroacupuncture frequency of 4/20 Hz for four weeks.After treatment,the rats were anesthetized,and bron-choalveolar lavage fluid(BALF)was collected for cell classification and counting.Lung tissues were collected for hema-toxylin-eosin(HE),and Masson's trichrome staining.The levels of IL-6,TNF-α,IL-1β,HO-1,SOD,and MDA in the BALF were measured.Western blot analysis was used to detect the expression levels of p-NF-κB,NF-κB,and I-κBαproteins in lung tissues.Results:Compared with the control group,the model group showed a significant increase in in-flammatory and oxidative stress-related factors(P<0.01),with HE and Masson's staining revealing collagen deposi-tion and alveolar structure disruption.Compared with the model group,the medication and electroacupuncture groups showed significant inhibition of NF-κB pathway phosphorylation(P<0.01),promotion of I-KBα expression(P<0.05),a significant reduction in the levels of IL-6,TNF-α,IL-1β,and MDA(P<0.001),and a significant increase in the levels of HO-1 and SOD(P<0.001).Additionally,electroacupuncture and medication reversed LPS-induced alveolar structural damage and collagen deposition.Conclusion:Electroacupuncture exerts therapeutic effects on COPD by inhibiting NF-κB protein phosphorylation,thereby showing potential as a treatment for COPD.
