摘要

Abstract

Objective To investigate the role of proline-glutamic acid-leucine-rich protein 1(PELP1)in angiogenesis of epithelial ovarian cancer.Methods Normal ovarian epithelial cells IOSE80 and epithelial ovarian cancer cells OVCAR-3 and Hey were selected.Hey-Ctrl group was set as blank control,Hey-OE-Vector group as negative control,and Hey-OE-PELP1 group as overexpression group.The relative expression level of PELP1 was detected by real-time fluorescence quantitative PCR(RT-qPCR)and Western Blot(WB).The relative expression level of PELP1 was detected by WB.The concentration of VEGFA in the supernatant(CM)of OE-Vector-CM group and OE-PELP1-CM group was determined by enzyme-linked immunosorbent assay(ELISA).The effects of CM on the migration and tube formation of human umbilical vein endothelial cells(HUVECs)were detected by Transwell and angiogenesis methods.Results The expression level of PELP1 in epithelial ovarian cancer cells OVCAR-3 and Hey was higher than that in normal ovarian epithelial cells IOSE80(P<0.05).The expression level of PELP1 in Hey-OE-PELP1 group was higher than that in Hey-Ctrl group and Hey-OE-Vector group(P<0.05).The concentration of VEGFA,the number of migrated cells and the number of tube nodes in OE-PELP1-CM group were higher than those in OE-Vector-CM group(P<0.05).Conclusion PELP1 promotes angiogenesis in epithelial ovarian cancer.
