摘要

Abstract

Objective To investigate the effect of neferine on renal oxidative stress in rats with diabetic nephropathy.Methods SD male rats were randomly divided into control group,model group,treatment group and positive control group.In addition to the control group,the other three groups were fed with high-sugar and high-fat diet to make DN model.After successful modeling,grouped drug treatment.The control group and the model group were intragastrically administered with the same amount of saline,the treatment group was intragastrically administered with 40 mg/kg neferine,and the positive control group was intragastrically administered with 14 mg/kg irbesartan.HE staining was used to observe the pathological conditions of rat kidney tissue under optical microscope.Biochemical analysis of urinary protein content in rats was performed.ELISA was used to detect the contents of TNF-α,SOD and MDA in rat kidney tissue.Results Compared with the control group,the renal tissue structure of the model group was significantly damaged,the urine protein of rats was significantly increased,the content of SOD in renal tissue was significantly decreased,and the contents of MDA and TNF-α were significantly increased(P<0.01).Compared with the model group,the local renal tissue of the treatment group was damaged to varying degrees,the urine protein of the treatment group was significantly reduced(P<0.01),the content of SOD was increased(P<0.05),and the content of MDA and TNF-α was decreased in the treatment group(P<0.05).Conclusion Neferine can improve the damage of renal tissue in diabetic nephropathy rats,reduce the content of urinary protein,and alleviate the oxidative stress level and inflammatory response in the kidney group of diabetic nephropathy rats.
