Abstract
Objective To investigate the influence of human papillomavirus(HPV)infection on the expression levels of human Runt-related transcription factor 3(RUNX3)and yeast gene Nob1 homolog(NOB1)in cervical cancer tissues of patients.Methods A total of 97 HPV-infected cervical cancer patients who underwent surgical treatment at Puyang Oilfield General Hospital from February 2021 to October 2023 were selected for the study.Additionally,30 patients with cervical intraepithelial neoplasia who underwent surgical treatment at the same hospital were selected as a control group.Based on gene amplification technique and the principle of flow-through hybridization,the results of hybridization between the amplified product and coated specific probe membrane were detected by reverse dot blot,and the status of HPV infection was qualitatively detected by alkaline phosphatase system.The expression levels of RUNX3 mRNA and NOB1 mRNA in the tissues were monitored in real-time using polymerase chain reaction to explore the relationship between virus type,viral load,and the expression levels of RUNX3 and NOB1.Results The expression level of RUNX3 mRNA in cervical cancer tissues was lower than that in cervical intraepithelial neoplasia tissues,while the expression level of NOB1 mRNA was higher than that in cervical intraepithelial neoplasia tissues(P<0.05).As the FIGO stage increased and with the occurrence of lymph node metastasis,the expression level of NOB1 mRNA increased and RUNX3 mRNA expression decreased,showing statistically significant differences(P<0.05).In patients with cervical cancer,those infected with high-risk HPV had lower RUNX3 mRNA expression levels than those with low-risk types,and the difference was statistically significant(P<0.05).Conversely,NOB1 mRNA expression levels were higher in patients with high-risk HPV infection than in those with low-risk types,with statistically significant differences(P<0.05).In patients with high HPV viral load,RUNX3 mRNA expression levels were lower than those with low viral loads,while NOB1 mRNA expression levels were higher(P<0.05).Spearman correlation analysis revealed a negative correlation between RUNX3 mRNA expression levels and viral load(r=-0.407,P<0.005),and a positive correlation between NOB1 mRNA expression levels and viral load(r=0.392,P<0.005).Logistic regression analysis indicated that high FIGO stage,lymph node metastasis,high-risk HPV infection,and high viral load were influencing factors for high expression of RUNX3 and NOB1.Conclusion In cervical cancer patients infected with HPV,the high expression of NOB1 and low expression of RUNX3 in cancer tissues are associated with disease staging and lymph node metastasis,and the type of HPV infection and viral load can influence the expression levels of NOB1 and RUNX3 in cancer tissues.关键词
