肉鸡屠宰及零售环节沙门氏菌污染的定性和定量检测方法比较
Comparison of Qualitative and Quantitative Detection for Salmonella Contamination in the Links of Broiler Slaughter and Retail

Abstract
In order to find out an optimal method for detecting the contamination rate and load of Salmonella in chicken,cotton swabs as well as washing liquor samples of carcasses/products were collected to compare the detection rate of Salmonella using cotton swab culture,carcass/product washing liquor culture,maximum probable number(MPN)method of carcass/product washing liquor and the combination of MPN method of carcass/product washing liquor and real-time fluorescent PCR(qPCR-MPN),respectively,and the loads of Salmonella were also compared using the last two methods.Qualitative detection results showed that the detection rate by qPCR-MPN method was the highest(84%),followed by MPN method of carcass/product washing liquor(80%),carcass/product washing liquor culture(60%)and cotton swab culture(28%),with statistically significant differences among the above methods(P<0.05);taking the carcass/product washing liquor culture as a standard,both qPCR-MPN and MPN method of carcass/product washing liquor were 100%sensitive and>80%consistent,but the latter was more specific.Quantitative detection results showed that more amount of Salmonella was detected by the qPCR-MPN method,compared to the MPN method of carcass/product washing liquor.In conclusion,the MPN method of carcass/product washing liquor was preferred for both qualitative and quantitative data;the qPCR-MPN method was highly consistent with the MPN method of carcass/product washing liquor,detecting more amount of Salmonella,but the risk of false positive should be noted when it was used for quantitative detection.关键词
