摘要

Abstract

To investigate the immunoregulatory effect of Trichinella spiralis C-type lectin(TsCTL)on mice,the TsCTL gene(GenBank accession number KRY42391.1)was cloned into the prokaryotic expression vector pET-28a(+)to construct the pET-28a(+)-TsCTL recombinant plasmid,which was expressed in Escherichia coli BL21(DE3).The recombinant TsCTL(rTsCTL)was purified,refolded,and identified by Western-blot,and endotoxin levels were measured.The rTsCTL was used to stimulate mouse spleen cells,and cell viability was assessed using the CCK-8 assay.The expression levels of transcription factors in the splenocyte were detected by qPCR.Ten C57BL/J mice were randomly divided into the control group(n=5)and the experimental group(n=5).Each mouse in the experimental group was subcutaneously injected with 50μg of rTsCTL(dissolved in 200 μL of PBS)three times a week for four weeks.The mouse in control group was treated with an equal volume of PBS.Splenocyte from both groups were extracted,and the levels of cytokines and transcription factors were measured using ELISA and qPCR.Finally,flow cytometry was used to assess the proliferation of regulatory T(Treg)cells.rTsCTL showed no toxicity to splenocyte.qPCR results indicated that rTsCTL could increase the expression levels of Gata3 and Foxp3 in splenocyte,while reducing the expression level of T-bet.The qPCR results from the animal experiments confirmed the same conclusion as those from the cell experiments.ELISA results showed that rTsCTL promoted the expression of TGF-β and IL-4,while inhibiting the expression of IFN-γ.Flow cytometry results demonstrated that rTsCTL increased the percentage of Treg cells.These findings suggest that rTsCTL can promote Th2 immune responses and inhibit Th1 immune responses.
