Abstract

To investigate the prevalence of Cryptosporidium infection in Tibetan sheep in high-altitude regions of Tibet,420 fresh fecal samples from Tibetan sheep were collected from intensive breeding farms in Gangba County,Shigatse City,Tibet,and Cryptosporidium infection was detected using nested PCR method to amplify the SSU rRNA gene;subtype analysis of the identified Cryptosporidium was conducted through analyzing the gp60 gene.The results showed that the total infection rate of Cryp-tosporidium in Tibetan sheep was 9.0%(38/420).SSU rRNA gene analysis revealed that the Cryptosporid-ium species infecting Tibetan sheep are Cryptosporidium xiaoi and Cryptosporidium ubiquitum.Statis-tical analysis revealed significant differences in the infection rates among different age groups and genders(P<0.05).Analysis of the gp60 gene showed that among the samples infected with C.xiaoi(n=22),18 samples were successfully classified into with 4 subtype families,including ⅩⅩⅢh(n=8),ⅩⅩⅢl(n=6),ⅩⅩⅢe(n=2),and ⅩⅩⅢd(n=2),whereas 16 samples with C.ubiquitum infection were all found to be the subtype Ⅻa(n=16).As Ⅻa subtype can infect humans and various other hosts,this data suggests a potential risk of zoonotic transmission of Cryptosporidium in sheep in this area,and effective control measures should be taken into consideration for this region.
