摘要

Abstract

The aim of this study was to investigate the effect of baicalin on the expression of typeⅠ interferon(IFN-Ⅰ)in the alveolar epithelial cells(MLE-12 cells)of mice infected with H9N2 avian influenza virus(H9N2 AIV),and further reveal the mechanism of baicalin's anti-avian influenza viral effects in vitro,as well as the relationship between baicalin and the immune function of the MLE-12 cells,with a view to providing theoretical basis for the antiviral mechanism of traditional Chinese medicine.A model of MLE-12 cells infected with H9N2 AIV was established in vitro,and the MOI of MLE-12 cells by H9N2 AIV was determined by plaque method,and the safe dose of baicalin was determined by toxicity assay,and the expression of IFN-Ⅰ in MLE-12 cells was detected by fluorescence quantitative PCR and ELISA.The MOI of MLE-12 cells infected by H9N2 AIV was 0.05,and the safe concentration of baicalein was less than 20 μg/mL.H9N2 AIV was able to induce the expression of IFN-α and IFN-β in MLE-12 cells.Its relative expression of IFN-α and IFN-β genes was significantly increased in baicalein high-dose group(15 μg/mL)at 24 h(P<0.05).The protein expression of IFN-α in MLE-12 cells was signficantly increased in the low,medium,and high dose groups of baicalin at 6 h(P<0.05),and the protein expression of IFN-β in MLE-12 cells was significantly increased at 12 and 24 h of action(P<0.05).H9N2 AIV induced the expression of IFN-αand IFN-β in MLE-12 cells.Baicalein could induce the expression of IFN-α and IFN-β in MLE-12 cells.Baicalin rapidly responds to H9N2 AIV infection by increasing the expression level of IFN-β in MLE-12 cells to exert antiviral effect.
