瑞香狼毒对小鼠肝脏损伤和糖代谢的影响
Effect of Stellera chamaejasme L.on Liver Injury and Glucose Metabolism in Mice

Abstract
To investigate the effects of the toxic plant Stellera chamaejasme L.on liver injury and glucose metabolism in mice,this study divided the mice into three groups:Negative control group,Experimental feeding I group,and Experimental feeding II group,which were fed 0,5,and 10 g/(kg·d)of Stellera chamaejasme L.,respectively.Clinical symptoms were observed,and body weight gain was measured.Serum biochemical parameters were analyzed using enzyme linked immunosorbent assay(ELISA),and liver histopathological changes were examined through hematoxylin-eosin(H.E.)staining.Quantitative real-time polymerase chain reaction(qPCR)was employed to assess the expression levels of glucose metabolism-related genes,including AMP-activated protein kinase α1(Ampkα1),signal transducer and activator of transcription 3(Stat3),glucose-6-phosphatase catalytic subunit 1(G6pc1),and sirtuin 1(Sirt1)in mouse liver.Western blot(WB)analysis was conducted to detect AMPKα1 protein expression in liver tissues.The results showed that both experimental groups exhibited varying degrees of depression,slow movement,and unkempt fur.Compared with the Negative control group,body weight gain in Experimental feeding I group was significantly reduced on days 7 and 28(P<0.05 or P<0.01),while in Experimental feeding II group,body weight gain was significantly reduced throughout the experiment(P<0.001).Additionally,body weight gain in Experimental feeding II group was significantly lower than in Experimental feeding I group on days 14,21,and 28(P<0.01 or P<0.001).Compared with the Negative control group,both experimental groups showed significantly reduced glucose(GLU)and total protein(TP)levels(P<0.001)and significantly increased alkaline phosphatase(ALP)and alanine aminotransferase(ALT)activities(P<0.001).In Experimental feeding II group,total cholesterol(TCH),total bilirubin(TBIL)levels,and aspartate aminotransferase(AST)activity were also significantly elevated(P<0.001).Compared with Experimental feeding I group,Experimental feeding II group exhibited significantly lower GLU levels(P<0.01)and significantly higher TCH,TBIL levels,and ALP,ALT,AST activities(P<0.01 or P<0.001).Histopathological examination revealed that both experimental groups exhibited structural disruption of hepatic cords,hepatocyte necrosis,congestion,and inflammatory cell infiltration.Compared with the Negative control group,Sirt1 and G6pc1 gene expression levels were significantly downregulated in Experimental feeding I group(P<0.05),while Ampkα1,Stat3,and G6pc1 expression levels were significantly downregulated in Experimental feeding II group(P<0.01 or P<0.001).The expression levels of Ampkα 1,Sirt1,Stat3,and G6pc1 genes in Experimental feeding II group were significantly lower than in Experimental feeding I group(P<0.05,P<0.01,or P<0.001).Furthermore,AMPKα1 protein expression in the liver was significantly lower in Experimental feeding II group than in the Negative control group and Experimental feeding I group(P<0.001).These findings indicate that Stellera chamaejasme L.induces pathological damage to liver tissue in mice,inhibits the expression of glucose metabolism-related genes and proteins,and disrupts hepatic glucose metabolism.关键词
