白杨素对糖尿病小鼠肝肾损伤的改善作用
Protective Effect of Chrysin on Liver and Kidney Injury in Diabetic Mice

Abstract
To investigate whether chrysin can improve liver and kidney damage in type 2 diabetic(T2DM)mice,40 male ICR mice were randomly divided into four groups:Control group,Model group,Metformin group,and Chrysin group.A T2DM mouse model was established by inducing a high-fat diet combined with intraperitoneal injection of streptozotocin(STZ)[150 mg/(kg·bw)].The Control and Model groups were administered physiological saline daily,the Metformin group was given metformin[200 mg/(kg·bw)],and the Chrysin group was given chrysin[100 mg/(kg·bw)]by oral gavage for 30 days.Fasting blood glucose(FBG)contents were measured every 6 days during the treatment period.After 30 days of treatment,blood was collected from the mice,and the liver,kidney,and pancreas were dissected.Serum contents of glycosylated serum protein(GSP),lipid profile[total cholesterol(TC),triglycerides(TG),low-density lipoprotein(LDL),and high-density lipoprotein(HDL)],malondialdehyde(MDA),creatinine(CRE),and urea were determined,along with the activity of aspartate aminotransferase(AST),superoxide dismutase(SOD),and alanine aminotransferase(ALT).Liver tissue was analyzed for the activity of SOD,glutathione peroxidase(GSH-Px),hexokinase(HK),pyruvate kinase(PK),and the content of MDA and liver glycogen.Hematoxylin-eosin(H.E.)stained pathological sections of liver,kidney,and pancreas tissues were examined for histological changes.The results showed that compared with the Control group,the Model group mice had significantly increased FBG contents,abnormal lipid metabolism,and a marked increase in serum GSP,TC,TG,and LDL contents(P<0.01),while HDL content and SOD activity were highly significant decreased(P<0.01).MDA contents were significantly higher(P<0.05),and ALT,AST activity,CRE,and urea contents were highly significant increased(P<0.01).SOD,GSH-Px,HK,PK activity,and liver glycogen content in liver tissue were highly significant reduced(P<0.01),while MDA contents were highly significant increased(P<0.01).Compared with the Model group,the Chrysin and Metformin groups showed improvements in lipid metabolism abnormalities.Both chrysin and metformin highly significant reduced serum GSP and MDA contents,AST activity,and CRE and urea contents(P<0.01),and highly significant increased SOD activity(P<0.01).Metformin highly significant decreased ALT activity(P<0.01).The activities of GSH-Px,HK,PK and liver glycogen content in liver tissue of both the Chrysin and Metformin groups were highly significant increased(P<0.01),and MDA contents were significantly reduced(P<0.05 or P<0.01).In addition,chrysin significantly increased SOD activity(P<0.05).Histopathological observations showed that both chrysin and metformin alleviated pathological damage in the liver,kidney,and pancreas of T2DM mice.The results indicate that chrysin has a certain protective effect on liver and kidney damage in T2DM mice.关键词
