Research Progress on Antiviral Effects and Molecular Mechanisms of Salicylamide Derivative Niclosamide

Abstract
Salicylamide-based drugs are widely used in both human and veterinary medicine due to their strong pharmacological activity and biological effects,particularly in the field of antiparasitic treatments.With the continuous advancement of the"drug repurposing"strategy,these drugs have garnered increasing attention in antiviral research.Niclosamide(NIC),a key representative of salicylamide derivatives,is primarily used to treat tapeworm infections and has demonstrated antibacterial,anticancer,and antiviral properties.Since the emergence of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)in 2019,researchers have screened over 3 000 Food and Drug Administration(FDA)-approved drugs and identified 83 potential anti-SARS-CoV-2 compounds,among which NIC exhibited a high selectivity index and was able to significantly inhibit viral replication at low concentrations.In addition to its inhibitory effects on SARS-CoV-2,NIC has shown potent antiviral activity against flaviviruses,adenoviruses,and herpesviruses.Mechanistically,NIC exerts its antiviral effects through multiple pathways,including modulating endosomal acidification,inhibiting syncytium formation,affecting intercellular signal transduction,regulating cellular metabolism,and inducing autophagy.This review summarizes the latest research progress on the antiviral properties and mechanisms of NIC,discusses its limitations,and explores potential strategies for optimization,aiming to provide new theoretical insights for antiviral drug development.关键词
