Study on Scalp Point through Point Acupuncture Regulating TWEAK/Fn14/STAT3 Signaling Pathway to Promote Angiogenesis in Rats Model of Intracerebral Hemorrhage

Abstract
Objective:To observe the effect of scalp point through point acupuncture on neuroethology and secondary cerebral edema in rats with intracerebral hemorrhage,and to explore the mechanism of its intervention in TWEAK/Fn14/STAT3 signaling pathway to promote angiogenesis.Methods:A total of 120 SD rats were randomly divided into the blank group,the sham operation group,the model group,the acupuncture group and the acupuncture+Stattic group,with 24 rats in each group.The intracerebral hemorrhage model was simulated by caudate nucleus autogenous arterial blood injection in rats.After the successful modeling,the acupuncture group and the acupuncture+Stattic group were treated with electro-needling Baihui(DU20)through Qubin(GB7),whereas the blank group,the sham operation group and the model group were not given any intervention.At 6 h,1 d,3 d and 7 d after the treatment,the Bederson score,Rosenberg score and brain water content were determined;the protein and mRNA expressions of TWEAK,Fn14 and STAT3 were detected by Western blot and Real-time PCR;the contents of MMP-2 and MMP-9 in hemotoma-side cortex were detected by ELISA.Results:The Bederson score,Rosenberg score and brain water content were significantly increased in the model group,the acupuncture group and the acupuncture+Stattic group compared with those in the blank group and the sham operation group at different time points(P<0.05);compared with those in the model group,the Bederson score,Rosenberg score and brain water content were significantly decreased in the acupuncture group on 1 d,3 d and 7 d after the intervention(P<0.05),in which there were no significant differences in the acupuncture+Stattic group at different time points(P>0.05).Compared with those in the acupuncture group,the Bederson score,Rosenberg score and brain water content were significantly increased in the acupuncture+Stattic group on 1 d,3 d and 7 d after the intervention(P<0.05).After the intervention,the protein and mRNA expressions of TWEAK,Fn14 and STAT3 were significantly increased in the acupuncture group compared with those in the model group and the acupuncture+Stattic group(P<0.05),whereas the contents of MMP-2 and MMP-9 were significantly decreased in the acupuncture group compared with those in the model group and the acupuncture+Stattic group(P<0.05);there was no statistical differences in the above indexes between the model group and the acupuncture+Stattic group(P>0.05).Conclusion:The intervention of intracerebral hemorrhage by scalp point through point acupuncture can significantly improve the neurobehavioral and secondary cerebral edema of rats,the mechanism of which may be through regulating TWEAK/Fn14/STAT3 signaling pathway to promote angiogenesis.关键词
