摘要

Abstract

The protective effect of piperine(PIP)on acute liver injury caused by carbon tetrachloride(CCl4)and the potential molecular mechanisms were explored.Forty male Kunming mice were stochastically divided into the normal control(NC)group,model(CCl4)group,positive control group treated with silymarin(SIY),low dose of PIP group(10 mg/kg CCl4+PIP)and high dose of PIP group(20 mg/kg CCl4+PIP).Pathological changes in livers,Liver index and liver function indices of all the mice were detected.While the activities or levels of SOD,GSH-Px,MDA,TNF-α,IL-1β,and IL-6 in the liver were respectively evaluated by relevant kit assays;Gene transcript levels and protein expression of NLRP3,ASC,Caspase-1 and IL-1β in the liver were measured using quantitative real-time PCR,Western blot,and immunohistochemistry.Results showed PIP effectively improved the indexes of liver function and pathological changes of liver tissue,alleviated the oxidative stress state.Further research showed that PIP reduced the mRNA and the protein expressions of NLRP3,ASC,caspase-1 and IL-1β.In conclusion,PIP may play a protective role in CCl4-induced acute liver injury by alleviating oxidative stress and inhibiting inflammatory reactions.
