Chemical Components and Antioxidant Effects of Water Extract from the Roots of Spuriopimpinella brachycarpa

Abstract
Objective:To study the antioxidant capacity of the water extract from the roots of Spuriopimpinella brachycarpa(Kom.)Kitag.and its effect on the lifespan of Caenorhabditis elegans,and to explore its medicinal potential.Methods:The chemical components in the water extract from the roots of S.brachycarpa were identified using ultra-high-performance liquid chromatography coupled with quadrupole Orbitrap mass spectrometry(UHPLC-Q-Exactive Orbitrap-MS).Wild-type and transgenic SOD-3::GFP C.elegans were used to evaluate the antioxidant effects of the water extract from S.brachycarpa roots.An oxidative stress model was induced by 100 mmol·L-1 paraquat,and different concentrations(125-500 mg·L-1)of the extract were administered to L1 stage C.elegans.The effects on antioxidant enzyme activity,reactive oxygen species(ROS)levels,and lifespan were evaluated.Results:Chemical analysis revealed that the water extract from the roots of S.brachycarpa contained phenolic acids such as chlorogenic acid,caffeic acid,and quinic acid,which exhibited significant antioxidant activity.The extract significantly reduced the production of ROS in the C.elegans,restoring them to normal levels,and rescued the decline in SOD-3::GFP expression caused by oxidative stress,thereby maintaining the activity of superoxide dismutase(SOD).The water extract also delayed the death of C.elegans induced by paraquat,showing strong antioxidant effects and prolonging lifespan.Conclusion:The water extract from the roots of S.brachycarpa enhances the antioxidant defense system of C.elegans,reduces oxidative stress,and prolongs lifespan,providing a scientific basis for its application as a natural antioxidant.关键词
