Mechanism of Action and Potential New Therapeutic Effects of Xiaoqinglong Decoction in Improving Respiratory Diseases Based on TPP-MS High-throughput Target Screening Technology

Abstract
Objective:By applying a targeted research method of classical prescriptions of traditional Chinese medicine(TCM)based on the research system of"classical prescriptions of TCM,high-throughput target screening,target association,and new clinical application",the mechanism of action of Xiaoqinglong Decoction(XQLD)in the treatment of pulmonary diseases was clarified,and its potential new therapeutic effects were explored.Methods:The technique of thermal proteome profiling(TPP)-high-resolution mass spectrometry(MS)was employed to perform high-throughput screening on the comprehensive targets of XQLD in rat lung tissue.The ingenuity pathway analysis(IPA)database was utilized to investigate the association between the targets of XQLD and downstream regulatory proteins.Furthermore,the Genevestigator platform was used for further exploration and analysis of target-associated diseases.Results:A total of 12 direct and 14 indirect targets of XQLD were screened.Through the combined analysis of the IPA database and the Genevestigator platform on the diseases associated with the targets of XQLD,it was found that these targets were involved in the treatment of bronchial asthma and chronic obstructive pulmonary disease and might treat allergic diseases and lung cancer through new anti-allergy and anti-tumor effects.Conclusion:This study clarifies the mechanism of action of XQLD in the treatment of pulmonary diseases from a target perspective and reveals its potential new therapeutic effects,including anti-allergic and anti-tumor properties.关键词
