基于指纹图谱结合网络药理学的山萸肉炮制前后差异质量标志物预测分析
Prediction and Analysis of Differential Quality Markers Before and After Processing of Cornus Officinalis Based on Fingerprinting Combined with Network Pharmacology

Abstract
Objective:To analyze the differential quality markers(Q-Markers)before and after the processing of Cornus Officinalis based on fingerprinting and network pharmacology techniques,and to establish a method for determining the content of differential Q-Markers.Methods:Ultra-performance liquid chromatography(UPLC)fingerprints of raw Cornus Officinalis,wine-processed Cornus,and honey-wine processed Cornus were established.Chemical pattern recognition methods were used to analyze the differential Q-Markers before and after processing.Network pharmacology techniques were employed to construct a"differential component-target-pathway"network to verify the rationality of these markers as potential differential Q-Markers,followed by quantitative analysis.Results:UPLC fingerprints of 10 batches of raw Cornus,wine-processed Cornus,and honey-wine processed Cornus identified 18,17,and 16 common peaks,respectively,with six chromatographic peaks identified using reference standards.The fingerprint similarity was greater than 0.9.Principal component analysis(PCA)identified three main factors.Orthogonal partial least squares discriminant analysis(OPLS-DA)parameters showed R2X=0.685,R2Y=0.843,and Q2=0.802,indicating good predictive ability and model fit.Based on the OPLS-DA model,the variable importance in projection(VIP)values ranked the peaks as follows:11,10,12,9,1,2,15,6,7,and 13.Quantitative results showed that the contents of 5-hydroxymethylfurfural,loganin,sweroside,cornuside I,and morroniside increased in honey-wine processed Cornus compared to wine-processed Cornus,while the content of gallic acid decreased.Based on the"five principles"of traditional Chinese medicine Q-Markers and network pharmacology analysis,gallic acid,loganin,sweroside,5-hydroxymethylfurfural,cornuside I,and morroniside were identified as differential Q-Markers before and after the processing of Cornus Officinalis.Conclusion:Fingerprinting,chemical pattern recognition,and network pharmacology techniques were used to preliminarily predict the differential Q-Markers before and after processing of Cornus Officinalis,providing a reference for quality evaluation and research on its pharmacological substance basis.关键词
