阿胶和阿胶肽对血管内皮细胞氧化损伤的作用研究
Study on Effects of Ejiao and Ejiao Peptide on Oxidative Injury of Vascular Endothelial Cells

Abstract
[Objective]To analyze the role of Corii colla asini(Ejiao)and Ejiao peptide in oxidative stress injury of vascular endothelial cells and the mechanism of their role.[Method]The model of human umbilical vein endothelial cell injury induced by hydrogen peroxide(H2 O2)was established.The effects of Ejiao and Ejiao peptide on the survival rate of human umbilical vein endothelial cells(HUVEC)before and after modeling were detected by CCK-8.The effects of Ejiao and Ejiao peptide on the activities of cell superoxide dismutase(SOD),glutathione per-oxidase(GSH-px),lactate dehydrogenase(LDH),and the contents of malondialdehyde(MDA),vascular endothelial growth factor(VEGF),interleukin-6(IL-6),nitric oxide(NO)were studied.[Result]Ejiao and Ejiao peptide could effectively promote the proliferation and migration of HUVEC,and could effectively resist the proliferation inhibition caused by oxidative injury.200 μg/mL Ejiao and Ejiao peptide could signifi-cantly inhibit the excessive production of MDA and LDH,200 μg/mL Ejiao could significantly increase the enzyme activity of GSH-px,Ejiao peptide could significantly increase the expression level of SOD,and repair the oxidative injury of HUVEC.Both Ejiao and Ejiao peptide could effectively promote the secretion of NO by vascular endothelial cells,protect vascular endothelial function,promote the secretion of VEGF by vascular endothelial cells,reduce the content of IL-6,promote angiogenesis,and reduce inflammatory reaction.[Conclusion]The mechanism of Ejiao and Ejiao peptide reducing oxidative stress of HUVEC may be related to improving cell growth and proliferation,enhancing cell antioxi-dant enzyme activity,promoting the expression of vascular endothelial growth factor and promoting the secretion of NO by vascular endothelial cells.关键词
