Optimization and establishment of tissue culture and rapid propagation technology system for Agave titanota'Red Catweazle'

Abstract
[Objective]Establish a tissue culture rapid propagation technology system of Agave titanota ′Red Catweazle′.[Method]The robust and the disease-free young plants of Agave titanota'Red Catweazle'were used as materials in our experiment,the effects of different base medium,disinfection schemes,and exogenous hormones on external plant disinfection,as well as the induction,proliferation,and rooting of the adventitious buds were studied;the effect of different cultivation substrates on the survival rate of tissue culture seedlings after transplantation were explored.[Result]The optimal disinfection treatment for the explants of Agave titanota'Red Catweazle'was a combination of 2%NaClO disinfection for 8 minutes and 0.1%HgCl2 solution disinfection for 12 minutes,with a infection rate of only 23.3%and a survival rate of 60.0%;using YMS as the base medium and adding 1.0 mg/L 6-BA were beneficial for the induction of Agave titanota'Red Catweazle',with an induction rate of 32.67%;adding 1.0 mg/L 6-BA,1.5 mg/L KT,and 0.5 mg/L NAA into YMS medium could promote the proliferation of adventitious buds,with a proliferation coefficient of 3.54;adding 1.0 mg/L IBA and 0.4 mg/L NAA into 1/2 YMS medium could effectively promote rooting,with a rooting rate of 85.6%and relatively healthy root growth;the survival rate of the adventitious buds of Agave titanota'Red Catweazle'after transplantation was 87.3%when using a mixture of peat soil,coconut coir and coarse sand with a ratio of 2∶2∶1.[Conclusion]Establishment of a tissue culture and rapid propagation technology system for Agave titanota'Red Catweazle'could provide technical references for achieving large-scale production and molecular breeding of Agave americana L.关键词
