Abstract

In order to explore the regulatory effect of wood vinegar on phosphorus conversion in kidney bean rhizosphere soil in saline-alkali habitat,and to determine the effect of wood vinegar application on the enzyme activity related to phosphorus conversion under saline-alkali stress,pot method was used in the experiment,and 5 different treatments were set up.The control group(CK)was treated with ultra-pure water and four different dilution groups(WV40,WV60,WV80,WV100)were treated with wood vinegar.After the first compound leaf of kidney bean was fully developed,the samples were taken to study the changes of phosphorus conversion related enzyme activity and gene abundance in kidney bean rhizosphere soil in saline-alkali habitat.The results showed that:the activities of alkaline phosphatase,acid phosphatase and phytase in rhizosphere soil were significantly higher than those in CK treatment,and the activities of alkaline phosphatase(ALP),acid phosphatase(ACP)and phytase in WV80 treatment all reached the maximum value.The activity of alkaline phosphatase,acid phosphatase and phytase was significantly increased by 15.18%,19.52%and 65.29%,respectively.The activity of neutral phosphatase in rhizosphere soil treated with CK,WV80 and WV100 was significantly higher than that treated with WV40 and WV60.The concentration of gene copy number of phytase coding gene(BPP)in WV80 treatment was the highest,and significantly higher than that in CK treatment,reaching 5.19×109 copies·g-1(soil).In alkaline phosphatase(phoD,phoX)coding genes,the gene copy number concentration of CK treatment was significantly higher than that of WV80 treatment.However,the concentration of acid phosphatase(phoC)gene copy number changed little in CK and WV80 treat-ment,and did not reach a significant level.The above results indicated that the activity of alkaline phosphatase,acid phosphatase and phytase could be improved by applying certain concentration of wood vinegar under saline conditions,but the activity of neutral phosphatase was not significantly improved.The copy number of BPP gene increased significantly under WV80 treatment,while the copy number of other phosphatase genes decreased.The results of this study were aimed at exploring the dynamic changes of the phosphatase in kidney bean rhizosphere soil under saline-alkali habitat,clarifying the activation of rhizosphere soil by wood vinegar,and providing basic data and theoretical support for the balance of saline-alkali habitat.
