多功能菌株不动杆菌的分离筛选鉴定及促生效果
Isolation,Screening and Identification of Multifunctional Strain Acinetobacter and Its Growth-promoting Effect

Abstract
Microbial strains with multiple functions were screened from the rhizosphere soil of wild Chinese sea buckthorn(Hippophae rhamnoides subsp.sinensis)in Qinghai,creating conditions for tapping rhizosphere growth-promoting strains with potential application value.The microorganisms in rhizosphere soil of Hippophae rhamnoides subsp.sinensis were isolated by selective culture media,and the strains were purified by streak plate method.The strains were identified through morphologi-cal,physiological and biochemical characteristics and 16S rDNA sequence comparison,and their abilities to dissolve organic phosphorus,inorganic phosphorus,potassium and degrade cellulose were determined to promote seed germination and seedling growth and development.The results showed that five strains of Acinetobacter were isolated from the rhizosphere soil of Hippophae rhamnoides subsp.sinensis.After three days of culture,the diameter of the turbid circle of the dissolved organic phosphorus was 5.97-8.85 mm,the diameter of the transparent circle of the dissolved inorganic phos-phorus was 2.56-6.75 mm,the amount of the dissolved organic phosphorus was 5.28-18.95 μg/mL,and the amount of the dissolved inorganic phosphorus was 3.63-21.52 μg/mL.The diameter of the yellow aperture for dissolving potassium was 7.37-25.13 mm,the amount of dissolving potassium was 2.92-8.60 μg/mL,and the diameter of the transparent circle for degrading cellulose was 9.62-11.15 mm.The results of plate growth promotion test showed that Acinetobacter could improve the germination rate of water spinach seeds and significantly improve the growth and development of wa-ter spinach seedlings.Among them,strain BD-1 had the best growth-promoting effect,and its leaf length and stem length were 18.28 mm and 62.98 mm,respectively,which were significantly in-creased by 17.4%and 76.9%compared with ck.After comprehensive evaluation,it was concluded that the growth status of water spinach was improved to some extent and the germination rate was sig-nificantly improved under the treatment of different Acinetobacter strains,among which BD-1 and BD-5 had better effects.关键词
