摘要

Abstract

Using Rhizoctonia solani as target pathogen,the optimal fermentation medium and shake flask fermentation conditions for the biocontrol bacterium Bacillus siamensis N2 were screened.Us-ing single factor level test and orthogonal test,the optimal fermentation medium formula and shake flask fermentation conditions were determined based on the growth rate and inhibitory rate of B.sia-mensis N2 against R.solani.The optimal fermentation medium formula for B.siamensis N2 is:glu-cose 20 g/L,peptone 50 g/L,glycerol 5 mL/L,K2HPO4·7H2O 1.5 g/L,and MgSO4·7H2O 1.5 g/L.The optimal conditions of shaker fermentation were:inoculation amount 4%,flask loading 100 mL/250 mL,culture time 24 h,culture temperature 28℃,rotation speed 180 r/min,and initial pH value 7.The study lays a theoretical foundation for the optimization of tank fermentation conditions and industrial production of N2 strain.
