Abstract

To enhance the purity of polysaccharides(LDP)extracted from the fruiting bodies of Lyophyllum decastes,a single-factor experiment was conducted as the foundation.The Box-Behnken central composite design principle was applied,with LDP content as the optimization indicator,to op-timize the conditions of three factors:solid-to-liquid ratio,extraction time,and disodium ethylenedi-aminetetraacetate(EDTA-2Na).The results showed that the highest LDP content of 75.47%was achieved when the solid-to-liquid ratio(g:mL)was 1∶22,the extraction time was 3 hours,and the EDTA-2Na addition was 1%.In the response surface optimization experiment,the factors affecting LDP content in descending order were solid-to-liquid ratio,extraction time,and EDTA-2Na.Analysis of monosaccharide composition and relative molecular weight revealed that the retention time of LDP was 10.117 minutes,with a relative molecular weight of 51 770 and a purity of 83%.The monosac-charide composition was primarily glucose and galactose,accompanied by a certain amount of man-nose,fucose,glucuronic acid,ribose,and a small amount of xylose.The findings of this study will provide a reference for further research on polysaccharides from Lyophyllum decastes.
