鹿皮胶制备工艺及增免活性研究
Preparation Technology of Deerskin Glue and Its Immunoenhancing Activity

Abstract
Deerskin was used as raw materials to prepare Manchu medicine deerskin glue,the or-thogonal experimental design method was employed to investigate the effects of steaming process on the yield and protein content of deerskin glue.The optimal steaming process for deer skin glue pro-duction was identified.Sixteen amino acids in steamed deerskin glue was determined.The immuno-modulatory effects of deerskin glue on normal mice were investigated by gavage administration,using thymus index,spleen index,and changes in tissue structure as indicators.Flow cytometry was em-ployed to detect the distribution changes of T lymphocyte subsets(CD4+and CD8+)in mice,and ELISA was used to measure the changes in cytokine levels(tumor factor-α,interferon-γ,interleukin-2,interleukin-4)in mice.The results showed that the steaming time of deerskin glue had a significant difference in the yield and protein content(P<0.01).The essential amino acid content in the deerskin glue obtained by steaming accounted for about 17.4%of the total amino acid content;deerskin glue increased the thymus index and spleen index of normal mice(P<0.05,P<0.01);The secretion of rat se-rum cytokines(IFN-γ,TNF-α,IL-2,IL-4)had a concentration-dependent promotion effect(P<0.05,P<0.01);The high-dose deerskin glue group can increase normally peripheral blood lymphocyte sub-sets CD4+ratio in mice(P<0.01),and each dose of deerskin glue can increase the value of peripheral blood CD4+/CD8+in mice(P<0.05,P<0.01);Deerskin glue did not cause significant abnormal patho-logical irritation or damage to the spleen tissue morphology of normal mice.The optimal steaming process for deer skin glue was determined to be at steaming pressure 0.1 MPa,steaming time 8 h,and power 800 W.The deerskin glue obtained through this process has the ability to modulate the im-mune function of normal mice.关键词
