阔叶箬竹竹叶中黄酮碳苷的提取及酸水解过程研究
Extraction and Acid Hydrolysis Process of C-glycosyflavone From the Leaves of Indocalamus latifolius

Abstract
Taking the leaves of Indocalamus latifolius as the raw material,the extract of I.latifolius leaves was initially obtained by the ultrasonic-assisted ethanol extraction method.Simultaneously,a high performance liquid chromatography(HPLC)method was established to analyze 4 flavonoid carbon glycosides(vitexin,isovitexin,orientin,and isoorientin)in the estracts of I.latifolius leaves.Then,the extract was hydrolyzed by acid and the effects of pH value,time,and temperature on the total content of 4 flavonoids were investigated.The chemical components of the leaves extracts before and after acid hydrolysis were fully characterized by HPLC-TOF-MS,and the chemical composition changes of the extract of I.latifolius leaves during acid hydrolysis were preliminarily explored.The results showed that the total mass fraction of 4 flavonoid glycosides in the leaves of I.latifolius was 0.92%.After hydrolysis at 80 ℃ and pH value of 2 for 1.5 h,the total mass fraction of 4 main flavonoid glycosides in the sample increased to 1.16%,which was 26.09%higher than that before hydrolysis.HPLC-TOF-MS detected 28 compounds,including 8 flavonoid carbosides,4 flavonoid oxysides,9 lignans,and 7 other compounds in the leaves extracts of I.latifolius.20 compounds could be detected in the samples after hydrolysis.Among them,the intensity of 8 flavonoid glycosides was significantly reduced and even undetectable by mass spectrometry,while the intensity of 4 main flavonoid carbon glycosides increased.In the process of acid hydrolysis,the oxyglycoside bond and carboglycoside bond were broken in the flavonoid glycoside compounds in the extract,and were most likely to be converted into isovitexin,thus promoting the increase of flavonoid glycoside content.关键词
