摘要

Abstract

Aim To investigate the effects of acupuncture combined with rehabilitation training on cerebral blood flow and the adenosine monophosphate-activated protein kinase(AMPK)/glutamine-fructose-6-phosphate aminotransferase-1(GFAT-1)/vascular endothelial growth factor(VEGF)signaling pathway in mice with ischemic stroke.Methods Sixty mice were randomly divided into sham,model,rehabilitation,acupuncture,and combined(rehabilitation+acupuncture)groups,with 12 mice in each group.Except the sham group,the other groups were subjected to ischemic stroke modeling.After 14 days of intervention,neuro-logical function was assessed using the Longa score,cerebral blood flow perfusion was measured by laser speckle imaging,brain tissue pathology was observed by hematoxylin-eosin(HE)staining,cerebral infarction volume was evaluated by triphenyltetrazolium chloride(TTC)staining,interleukin(IL)-6,IL-1β,and tumor necrosis factor-α(TNF-α)levels were detected by enzyme-linked immunosor-bent assay,and AMPK,phosphorylated AMPK(p-AMPK),GFAT-1,and VEGF protein levels were measured by Western blotting.Results Compared with the sham group,the Longa score,cerebral infarction volume,IL-1 β,IL-6,TNF-α,and GFAT-1 protein lev-els were increased,while cerebral blood flow perfusion,AMPK,p-AMPK,and VEGF protein levels were decreased in the other groups(P<0.05).Compared with the model group,the rehabilitation,acupuncture,and combined groups showed significant improvements in these indicators,with the combined group showing the most significant improvement(P<0.05).The model group exhibited disor-dered cell arrangement,enlarged intercellular spaces,and necrosis,while the rehabilitation,acupuncture,and combined groups showed significant pathological improvements,with the combined group showing the best results.Conclusion Acupuncture com-bined with rehabilitation training can improve neurological damage and protect brain tissue in mice with ischemic stroke by regulating the AMPK/GFAT-1/VEGF signaling pathway.
