基于PI3K信号通路探讨麝香促进骨髓间充质干细胞迁移的作用机制
Exploring the mechanism of musk in promoting migration of bone mesenchymal stem cells through the PI3K signaling pathway

Abstract
Objective To investigate whether the phosphatidylinositol-3-kinase(PI3K)signaling pathway plays a role in the musk-promoted migration of bone mesenchymal stem cells(BMSCs).Methods Sixty SD rats were randomly divided into low/medium/high-dose musk groups and the blank control group,and medicated serum was prepared.Fifteen SD rats were used to isolate and culture BM-SCs.The third-generation BMSCs were identified by morphology,flow cytometry,osteogenic and adipogenic induction.The third-gen-eration BMSCs were then treated with different doses of musk medicated serum,and the cell proliferation rate was detected by MTT as-say.Then the effect of musk medicated serum on the migration ability of BMSCs in vitro was observed by adding the PI3K signaling path-way inhibitor(LY294002).Results The isolated cells exhibited characteristic MSCs morphology.Phenotypic identification showed posi-tive expression of CD44 and CD90,but no expression of CD45 or CD34.The cells could be induced to differentiate into osteogenic and adipogenic cells.Compared with the control group,the proliferation rate of MSCs of all dose musk groups were significantly increased on days 1-7(P<0.05).Transwell migration assays revealed that low-dose musk medicated serum significantly promoted BMSCs migra-tion at 24,48,and 72 h(P<0.05).After adding the inhibitor LY294002,the number of migrating cells were decreased significantly at different time points(P<0.05).Conclusion Low-dose musk medicated serum significantly enhances BMSCs migration,and its mech-anism may be associated with the PI3K signaling pathway.关键词
