基于PI3K/AKT信号通路的右归丸防治悬尾骨质疏松症大鼠的机制研究
Study on the mechanism of Yougui Pill(右归丸)in preventing and treating tail-suspended osteoporosis in rats based on the PI3K/AKT signaling pathway

Abstract
Objective To observe the effects of Yougui Pill(右归丸,YGP)on the PI3K/AKT signaling pathway in the bone tissues of rats with tail-suspended osteoporosis(OP)and elucidate the intrinsic mechanism of YGP in preventing and treating OP.Methods An OP rat model was established by tail suspension for 8 weeks,with simultaneous administration of YGP via gavage.After gavage,the con-tents of alkaline phosphatase(ALP)and tartrate-resistant acid phosphatase(TRACP)in the abdominal aorta serum were measured u-sing ELISA.Pathological changes in the distal left femur were observed via hematoxylin-eosin(HE)staining.Bone mineral density(BMD)of the left femur was assessed using dualenergy X-ray absorptiometry(DXA).Micro-computed tomography(Micro-CT)was employed to analyze the morphological changes in the distal left femur,along with bone volume fraction(BV/TV),trabecular thick-ness(Tb.Th),and trabecular separation(Tb.Sp).Western blot(WB)was used to detect protein expressions of PI3K,p-PI3K,AKT and p-AKT in the femoral tissues.Results Compared with the blank group,the model group exhibited decreased expressions of ALP,BMD,BV/TV,Tb.Th,p-PI3K/PI3K,and p-AKT/AKT,while the TRACP level was elevated(P<0.01).HE staining and Micro-CT revealed that,compared with the blank group,the model group showed reduced and fractured trabeculae,thinner trabeculae,increased spacing,and decreased trabecular area.These changes were ameliorated following YGP intervention.Conclusion YGF can enhance the expression of PI3K/AKT signaling pathway in the bone tissues,and improve the bone microstructure in tail-suspended OP rats,thus playing its role in preventing and treating OP.关键词
