摘要

Abstract

Objective To investigate the mechanism of Tangduqing(糖毒清,TDQ)in ameliorating insulin resistance in type 2 diabetic rats through the aryl hydrocarbon receptor(AHR)/interleukin-22(IL-22)signaling pathway.Methods Sixty SDrats were divided into the normal control group(n=10)and model group(n=50).Type 2 diabetic models were established with 50 rats by intraperitone-al injection of streptozotocin(STZ).The 44 successfully modeled rats were randomly and equally divided into the model group,metform-in hydrochloride group,TDQ low-dose group,and TDQ high-dose group.After six weeks of continuous gavage,the area under the curve(AUC)was tested using the oral glucose tolerance test(OGTT),insulin resistance was evaluated using homeostasis model assess-ment of insulin resistance(HOMA-IR)and quantitative insulin sensitivity index(ISI).Indole contents were measured via liquid chro-matograph mass spectrometer(LC-MS).Serum levels of IL-22 and IL-6 were detected by ELISA.Immunohistochemistry and HE staining were used to assess the intestinal tissues and the expressions of AHR and IL-22.Results TDQ reduced blood glucose,fasting insulin(FINS),AUC,and HOMA-IR(P<0.05)while increased ISI(P<0.05)in STZ-induced diabetic rats.It elevated bacteri-al metabolite indole levels,suppressed IL-6 level in serum(P<0.05),enhanced IL-22(P<0.05),alleviated intestinal inflamma-tory infiltration,and up-regulated AHR and IL-22 expression in the intestinal tissues.Conclusion TDQ ameliorates insulin resistance and suppresses metabolic inflammation,and its mechanism may be associated with the activation of the AHR/IL-22 inflammatory path-way.
