Abstract

Objective To investigate the effects of modified Gexia Zhuyu Decoction(膈下逐瘀汤,GXZYD)on the the tissue structure of fallopian tubes in salpingitis-induced infertility(SOI)rats with qi stagnation and blood stasis based on the TLR4/MyD88/NF-κB/NLRP3 signaling pathway.Methods A disease-syndrome combined model of chronic SOI was established.After intervention with modi-fied GXZYT and the TLR4 pathway inhibitor TAK-242,plasma,serum,and fallopian tube tissues were collected.Serum levels of in-terleukin-1β(IL-1β)and tumor necrosis factor-α(TNF-α)were measured by enzyme-linked immunosorbent assay(ELISA).Hemorheological parameters were assessed to evaluate the state of qi stagnation and blood stasis.Pathological changes in fallopian tubes were observed by hematoxylin-eosin(HE)staining.Immunohistochemistry(IHC)was used to detect the expressions of TLR4 and NL-RP3.RT-Pcr and Western blot(WB)were performed to analyze mRNA and protein expression levels of TLR4,MyD88,NF-κB,NLRP3,Bax,and Bcl-2.Pregnancy rates were observed.Results Compared with the model group,the modified GXZYT group showed significantly reduced plasma viscosity andwhole bloodviscosity(3.0 s-1).Bothgroups exhibiteddecreasedserum IL-1βand TNF-α levels,down-regulated mRNA and protein expressions of TLR4,MyD88,NF-κB,NLRP3 and Bax,and up-regulated Bcl-2 mR-NA and protein expressions.Pathological status in fallopian tubes was alleviated,and pregnancy rates were increased in both groups.Conclusion Modified GXZYT alleviates chronic SOI with qi stagnation and blood stasis by modulating the TLR4/MyD88/NF-κB/NL-RP3 pathway,reducing apoptosis and inflammation.TAK-242 can play the role of modified GXZYT to improve inflammatory indexes and pathological state of the disease,but does not alleviate syndrome manifestations of traditional Chinese medicine(TCM).
