二仙汤对去势高血压大鼠血管内皮舒张功能与GPER信号的影响
Effects of Er'xian Decoction(二仙汤)on vascular endothelial diastolic function and GPER signaling in castrated hypertensive rats

Abstract
Objective To investigate the effects of Er'xian Decoction(二仙汤,EXD)on improving vascular endothelial function and its impact on G protein-coupled estrogen receptor(GPER)signaling in castrated hypertensive rats.Methods Ovariectomized female spon-taneously hypertensive rats(SHR)were divided into the model group(0.9%saline),EXD group(15.12 g/kg),positive control group(captopril,1 mg/kg),and control group(ovariectomized WKY rats,0.9%saline),with 6 rats in each group.All groups received in-tragastric administration for 4 weeks.After 4 weeks,systolic blood pressure(SBP),diastolic blood pressure(DBP),and heart rate(HR)were measured.Aortic tissues were analyzed using Masson staining,Elastica van Gieson staining,and transmission electron mi-croscopy(TEM).Levels of nitric oxide(NO),endothelin-1(ET-1),and angiotensin II(Ang II)in the aortic tissues,as well as serum estradiol(E2),matrix metalloproteinase-2(MMP-2),desmosine(DES),endothelial nitric oxide synthase(eNOS),cyclic guanosine monophosphate(cGMP),and NO were detected using chemical assays or ELISA.Western blot(WB)was used to measure protein expression levels of MMP-2,collagen III,GPER,eNOS,and phosphorylated eNOS(p-eNOS)in the aortic tissues.Re-sults Compared with the control group,the model group exhibited elevated blood pressure,significant endothelial structural changes,de-creased serum contents of E2,NO,eNOS,and cGMP,increased contents of MMP-2 and DES,reduced NO content and GPER,eNOS,and p-eNOS protein expression in tissues,and increased ET-1,Ang II,MMP-2,and collagen III protein expression.Com-pared with the model group,the EXD group showed varying degrees of improvement in these indicators.Conclusion EXD may activate the expression of GPER in castrated hypertensive rats,promote eNOS-mediated NO secretion to enhance vasodilation,ameliorate vascu-lar endothelial fibrosis,and ultimately reduce blood pressure.关键词
