化瘀通络中药通过Hedgehog信号通路对糖尿病大鼠肾脏纤维化的干预作用研究
Study on the interventional effect of Huayu Tongluo Formula(化瘀通络方)on renal fibrosis in diabetic rats via the Hedgehog signaling pathway

Abstract
Objective To investigate the mechanism of Huayu Tongluo Formula(化瘀通络方,HYTLF)in intervening renal fibrosis in diabetic rats via the Hedgehog signaling pathway.Methods 10 rats were randomly selected as the control group from 60 male SD rats,and the rest were fed with high-sugar and high-fat diet combined with streptozotocin(STZ)to establish the diabetic model.The dia-betic rats were then randomly divided into the model group,irbesartan group,and HYTLF group.After 16 weeks of intervention,24 h u-rine and blood samples were collected to detect urine protein,fasting blood glucose(FBG)and renal function.Renal tissues were exam-ined by light and electron microscope.The mRNA and protein expressions of Shh,Gli1 and FSP1 in the renal tissues were detected by RC-PCR and Western blot(WB).Results Compared with the model group,both treatment groups showed significant reductions in 24 h urinary protein(P<0.01),with the HYTLF group outperforming the irbesartan group.Serum creatinine(Scr)and uric acid(UA)levels were decreased significantly in the HYTLF group(P<0.01).Renal pathological damage was alleviated in both treatment groups compared to the model group.The mRNA and protein expression contents of Shh,Gli1,and FSP1 were reduced in both treatment groups(P<0.05 or P<0.01),with the HYTLF group showing greater efficacy than the irbesartan group.Conclusion HYTL can reduce urina-ry protein excretion and protects renal function in diabetic nephropathy rats.Its mechanism may involve inhibiting the activation of the Hedgehog signaling pathway,thereby mitigating renal injury and delaying the progression of renal fibrosis.关键词
