托里消毒散对高糖诱导损伤M2型巨噬细胞分泌抗炎因子的影响
Effect of Tuoli Xiaodu Powder on the Secretion of Anti-inflammatory Factors in M2 Macrophages with Injury Induced by High Glucose

Abstract
Objective:To investigate the effect of Tuoli Xiaodu Powder on the secretion of anti-inflammatory and tissue repair fac-tors in M2 macrophages damaged by high glucose induction.Methods:Human monocytic THP-1 cells were induced to differentiate into M2 macrophages in vitro using phorbol 12-myristate 13-acetate(PM A),interleukin-4(IL-4),and interleukin-13(IL-13).The induced cells were identified by observing their morphology and detecting the expression of M2 macrophage surface markers argi-nase-1(Arg-1)and CD206 via Western blot.Successfully identified M2 macrophages were exposed to 33 mmol/L glucose for 120 h and then randomly divided into an experimental group and a model group.The experimental group was treated with 500 μg/mL Tuoli Xiaodu Powder,while the model group received an equivalent volume of phosphate-buffered saline(PBS).A normal group was also established,receiving the same volume of PBS as an intervention.After 48 h of intervention,the supernatants of the cell cul-tures were collected,and enzyme-linked immunosorbent assay(ELISA)was used to measure the absorbance of interleukin-10(IL-10),IL-4,IL-13,transforming growth factor-β 1(TGF-β1),vascular endothelial growth factor(VEGF),hepatocyte growth factor(HGF),insulin-like growth factor-1(IGF-1),platelet-derived growth factor(PDGF),and fibroblast growth factor(FGF).Results:Microscopy and Western blot analysis confirmed the successful polarization of THP-1 monocytes into M2 macrophages.ELISA results showed that compared to the normal group,the model group exhibited significantly lower levels of IL-10,IL-4,IL-13,VEGF,HGF,IGF-1,TGF-β1,PDGF,and FGF(P<0.05).Compared to the model group,the experimental group showed significantly increased lev-els of IL-10,IL-4,IL-13,VEGF,HGF,IGF-1,PDGF,and FGF(P<0.05),while the difference in TGF-β1 levels was not statistically significant.Conclusion:Tuoli Xiaodu Powder promotes the secretion of anti-inflammatory and tissue repair factors in M2 macrophages damaged by high glucose induction,suggesting its potential application in the treatment of diabetic skin ulcer wounds.关键词
