隔药灸脐法对失眠大鼠的治疗作用及机制
Therapeutic Effect and Mechanism of Herb-partitioned Moxibustion at the Navel in Insomnia Rats

Abstract
Objective:To investigate the effects of herb-partitioned moxibustion at the navel on the hypothalamic tissue morphology and peripheral blood levels of corticosterone(CORT),prostaglandin D2(PGD2),and interleukin-1 β(IL-1β)in rats with insomnia,and to explore its mechanism in improving insomnia.Methods:Twenty-seven male Sprague-Dawley(SD)rats were randomly divided into three groups according to a random number table:blank group,model group,and treatment group,with 9 rats in each group.The insomnia model was induced in the model and treatment groups by intraperitoneal injection of P-chlorophenylalanine(PCPA)using a modified method.Model establishment method:The PCPA solution was prepared in weakly alkaline physiological saline and adminis-tered by intraperitoneal injection at 0.5 g/kg for 2 consecutive days.After model establishment,the blank group was treated with e-qual volume of physiological saline and no further intervention.The model group received fixation of four limbs without additional in-tervention,while the treatment group received herb-partitioned moxibustion at the navel once every 2 days for a total of 10 treatments.After the intervention,rats were anesthetized by intraperitoneal injection with pentobarbital sodium,and blood was collected from the abdominal aorta.The hypothalamus was isolated after heart perfusion.The general condition of the rats was observed,and sleep was as-sessed by the pentobarbital sodium sleep test.Hematoxylin-eosin(HE)staining was used to observe the morphology of the hypothala-mus,and enzyme-linked immunosorbent assay(ELISA)was used to detect the levels of CORT,PGD2,and inflammatory mediator IL-1β in serum.Results:Compared to the model group,the general condition of the treatment group improved to varying degrees.Before the intervention,the model and treatment groups had lower body weight than the blank group(P<0.05).After the intervention,the body weight of the model group was lower than that in the blank group(P<0.05),and the treatment group was higher than the model group(P<0.05).Compared with the results before intervention,the body weight in the model group was decreased(P<0.05),and the body weight in the blank group and the treatment group was increased(P<0.05).After modeling,the sleep latency was longer(P<0.05)and the sleep time was shorter(P<0.05)in the model group and the treatment group compared to that in the blank group.After the intervention,compared to the blank group,the sleep latency in the model group and the treatment group remained lon-ger(P<0.05),and sleep time remained shorter(P<0.05).After intervention,compared to the blank group,the sleep latency in the model group was longer(P<0.05),and the sleep time was shorter(P<0.05).Compared to the model group,sleep latency was shor-ter(P<0.05)and sleep time was longer(P<0.05)in the treatment group.In the model group,compared to the results before mod-eling,sleep latency was longer(P<0.05)and sleep time was shorter(P<0.05)after modeling and intervention.In the treatment group,compared to the results before modeling,sleep latency was longer(P<0.05)and sleep time was shorter(P<0.05)after mod-eling,while compared to the results after modeling,sleep latency was shorter(P<0.05)and sleep time was longer(P<0.05).HE staining showed that,compared to the model group,the treatment group had darker nuclear staining,increased cell density,and more orderly arrangement of hypothalamic cells,which was similar to the blank group.The serum CORT level was higher in the model group compared to the blank group(P<0.05),but lower in the treatment group compared to the model group(P<0.01).The serum PGD2 level showed no significant difference between the model group and the blank group(P>0.05),but was significantly higher in the treatment group compared to the model group(P<0.01).The serum IL-1 β level was higher in the model group compared to the blank group(P<0.05),but lower in the treatment group compared to the model group(P<0.01).Conclusion:Herb-partitioned moxibus-tion at the navel can significantly improve the general condition,sleep quality,and hypothalamic tissue morphology in rats with insom-nia.It regulates the serum levels of CORT,IL-1β,and PGD2,and its mechanism may be related to hormone levels and inflammation.关键词
