Abstract
Three strains of H5 subtype HPAIV(Hn/Y4,Hn/SMQ9,and Hn/SMQ2)isolated from dead whooper swans in Henan and Shaanxi provinces,China,in 2016,2020,and 2021,respectively,were used in this study.Tissue samples(trachea,lungs,spleen,liver,and rectum)were collected from the swans.The viral load in each tissue was measured using real-time fluorescence quantitative PCR.The results showed that the viral load in the lungs and trachea was significantly higher than in other tissues,while the viral load in the rectum was lower.This distribution pattern suggests that H5 subtype HPAIV primarily relies on respiratory tissues(lungs and trachea)for efficient replication in whooper swans,with transmission likely occurring through respiratory secretions(such as droplets),which differs significantly from the fecal excretion mode in poultry.As a migratory waterfowl,the whooper swan's habitat and migration path may act as key nodes for cross-regional viral transmission,increasing the risk of contact with other birds or poultry.This study provides important scientific evidence for understanding the host tropism,transmission mechanisms,and prevention and control strategies for H5 subtype avian influenza virus.关键词
